The effects of saffron supplementation on the relation between HSP and cytokines following strenuous physical activity in male mountain climbers

Abstract
Introduction: The aim of this study was to evaluate the relation between Heat Shock Protein 70 (HSP-70), Interleukin-6 (IL-6) and Tumor Necrosis Factor-α (TNF-α) in one bout of strenuous physical activity with and without saffron supplementation in male mountain climbers.
Methodology: Participants were eleven male mountain climbers with 27-37 years of age. The physical activity session included running on a treadmill with %-5 gradient for 45 minutes with %80-90 maximum heart rate. Blood samples were taken in three different times- before, immediately and one hour after the physical activity- in order to measure HSP-70, IL-6 and TNF-α. After three days of recovery, participants started consuming 300 mg/d capsulated dried saffron stigma for 10 days. The physical activity session was repeated and blood samples were again taken before, immediately and one hour after it. Pearson's chi-squared test was used for analysis of results (α= 0.05).
Results: The physical activity protocol resulted in an increase in studied pro-inflammatory cytokines and HSP70 both before and after supplementation. Before the first physical activity session and supplementation, positive and considerable correlation was shown between IL-6 and TNF-α. Despite the relationships in other stages, none of them were statistically considerable.
Before consumption of saffron capsules, TNF-α and IL-6 had negative considerable correlations immediately and one hour after physical activity. Both IL-6 and TNF-α decreased after one hour of physical activity, but IL-6 decreased more than TNF-α and they had contrary concentrations, meaning the more IL-6 decreased, TNF-α showed less decrease. Further results show that TNF-α and HSP70 decreased immediately and one hour after physical activity both before and after supplementation and their trends were positively and significantly correlated.
Discussion:Ten days Saffron supplementation showed positive influences on mentioned immune responses after physical activity. Due to supplement’s antioxidant and anti-inflammatory ingredients,a positive correlation between TNF-α and HSP-70 happened immediately and one hour after physical activity.
